Totally black screen after turning it on, what could be the issue?
I have an 2011 iMAC 27 desktop, recently a totally black screen shows up after turning it on, I can hear the chime sound, but the screen is totally black. Can anyone advise what could be the issue? And how to isolate it?

Using a bright flashlight rest it on the display on the running system as a sharp angle are you able to see the faint image of your desktop icons? An alternative way would be to connect an external display via the miniDP connection to the Thunderbolt port in the back.
One other test would be to connect a second mac system via a network connection are you able to access it remotely? This would isolate out the issue as being a display only problem.

Thanks for the quick response, Dan. I gave it a try, 4 times, and found the screen can light up the first two times, but completely dark at the rest. If I connect it to an external monitor through display port, should I remove the screen display or can have it attached? Any thoughts?

What do you mean by 'light up' are you trying to say the backlight is working, just not seeing an image? If thats so, the issue is within the GPU logic, not the backlight. You'll need a new GPU board to fix your system as @mayer pointed out.
Your system is designed to support two displays, no need to disable the internal.

I mean the display is working in the first 2 times, I can see images, but completely dark when I try the 3rd and 4th times (full shutdown and turn it on).
