Should you not be able to hear anyone while on handset mode, but hear people on loudspeaker mode, you would need to replace your earspeaker.
should you not hear on either mode, then both speakers could possibly be blown, though there could be an issue with your audio controller IC.
there is a possibility that a factory reset could fix your issue as data corruption can cause this issue, but is less common.
